The Evidence

Early Phase 2 signal (12-week trial)

An earlier, smaller trial reported a modest weight-loss difference versus placebo (2.6 kg vs 0.8 kg) that generated initial optimism about the compound.

Pivotal Phase 2b trial (536 participants, reported 2007)

In the larger, 24-week OPTIONS trial with oral dosing, AOD-9604 failed to produce statistically significant weight loss versus placebo. Development for the obesity indication was terminated, and the company never advanced it to Phase 3 or published the negative results in a peer-reviewed journal.

Honest limits

Some researchers have suggested the oral delivery method — rather than the underlying biology — may explain the failure, since peptides generally degrade in the GI tract, but no large injectable trial was ever run to test that. AOD-9604 has never received FDA approval as a drug for any indication and has no legitimate U.S. compounding pathway.
