Multiple randomized, placebo-controlled trials of oral NAD+ precursors (nicotinamide riboside, nicotinamide mononucleotide) in middle-aged and older adults consistently show the supplements are well tolerated and reliably raise circulating NAD+ levels.
Functional outcome trials — mixed to limited
The same trials have generally not shown clear improvements in cardiovascular, cognitive, or muscle-strength outcomes; a 2025 systematic review found human effects on metabolic and performance outcomes remain mixed, with small sample sizes and short durations as recurring limitations.
IV/injectable-route trial data
A 2025 PRISMA-guided systematic review found no eligible outcomes trials specifically testing IV or IM NAD+ for anti-aging or wellness purposes — the direct-injection route used in many clinics has less formal trial evidence behind it than oral precursor supplementation.
Animal research
Preclinical studies in rodents show more consistent benefits for metabolism, inflammation, and mitochondrial function, but that evidence has not yet reliably translated into measured human functional gains.
