The Evidence

Early small human trials (1980s)

Studies with as few as 6 to 16 participants — including chronic-insomnia patients — reported improved sleep efficiency, shorter sleep latency, and longer sleep duration with IV DSIP versus placebo, without daytime sedation.

Contradicting well-controlled studies

Other well-controlled trials from the same era found no statistically significant hypnotic effect — DSIP did not reliably increase total sleep time or slow-wave sleep compared with placebo, making inconsistency the defining feature of the human research record rather than an edge case.

No confirmed receptor or gene

Despite roughly two decades of active study, researchers have never conclusively identified a DSIP receptor or gene, which is part of why scientific interest declined and few new controlled human trials have been published in the last twenty-plus years.

Conflicting hormonal data

Human studies on DSIP's effect on stress hormones directly conflict — one study found reduced circulating ACTH, another found no effect on CRH-stimulated ACTH and cortisol — underscoring how unresolved the basic pharmacology remains.
