KPV is a tripeptide fragment — lysine-proline-valine — derived from alpha-melanocyte-stimulating hormone (alpha-MSH). It retains alpha-MSH's anti-inflammatory activity without the pigmentation effects, and is studied mainly in gut-inflammation research models.
Unlike full-length alpha-MSH, KPV does not require a functional melanocortin receptor to act — it appears to work intracellularly, inhibiting NF-κB and MAP-kinase inflammatory signaling directly, and is transported into intestinal cells via the PepT1 transporter, which is notably upregulated during intestinal inflammation.

KPV has reduced disease activity, inflammatory cytokine levels, and tissue-injury scores in both DSS- and TNBS-induced colitis in mice, including when given orally — suggesting gastrointestinal stability that's notable for a peptide, though this is animal, not human, evidence.
